Assuming you download the same file, no.
Some downloaders will try to get the highest quality available instead of something that's "reasonable".
Either way though you don't have to stream it multiple times if you really like it, so if it's a video you plan to view many times it will for sure save some bandwidth.
Also lets you keep it in case youtube goes retarded and takes it off for silly copyright claims.

Youtube actually downloads the file when you stream it. :P It then deletes the file when you no longer have it open.

Downloading files is usually much higher quality vs. streaming. Started watching Bakemonogatari recently, and streaming quality fails in comparison to the 480p downloads I get, which are linked in the page where I'm streaming from.
